如何完成一次又經(jīng)濟(jì)的Tunel檢測(cè)?
——Yeasen TUNEL detection Kit 讓“美”原位呈現(xiàn)
通過(guò)TdT轉(zhuǎn)移酶將Fluorescein-dUTP結(jié)合到有缺口的DNA上是檢測(cè)細(xì)胞凋亡的常用方法之一,如Roche提供的In Situ Cell Death Kit正是基于這個(gè)原理。產(chǎn)品中的TdT轉(zhuǎn)移酶活性直接影響到標(biāo)記的效率。翊圣生物立足于酶制劑生產(chǎn),力求開(kāi)發(fā)以酶制劑為基礎(chǔ)的產(chǎn)品,推出三款熒光素(Alexa Fluor 640,Alexa Fluor 488,FITC)標(biāo)記的Tunel檢測(cè)試劑,在滿足客戶對(duì)產(chǎn)品高品質(zhì)要求的同時(shí)又可以享受低至Roche一半的價(jià)格。

檢測(cè)機(jī)理:
細(xì)胞凋亡晚期,染色體DNA雙鏈斷裂或單鏈斷裂產(chǎn)生大量的粘性3-OH末端,在脫氧核糖核苷酸末端轉(zhuǎn)移(TdT)的作用下,將熒光素/酶標(biāo)記的dUTP結(jié)合到DNA的3-末端,從而可通過(guò)檢測(cè)熒光完成對(duì)細(xì)胞凋亡的檢測(cè),這類(lèi)方法稱(chēng)為脫氧核糖核苷酸末端轉(zhuǎn)移酶介導(dǎo)的缺口末端標(biāo)記法(Terminal-Deoxynucleotidyl Transferase Mediated Nick End Labeling,TUNEL),原理見(jiàn)圖1。

產(chǎn)品特點(diǎn):
適用范圍廣:可用于石蠟包埋組織切片、冰凍組織切片、培養(yǎng)的細(xì)胞和從組織中分離的細(xì)胞。
檢測(cè)靈敏度高:可檢測(cè)出極少量的凋亡細(xì)胞。

常見(jiàn)問(wèn)題:
Q:出現(xiàn)非特異性熒光標(biāo)記?
1)組織/細(xì)胞本身核酶或聚合酶活性水平較高,易導(dǎo)致出現(xiàn)非特異性的熒光標(biāo)記,例如平滑肌細(xì)胞。解決方法是,取細(xì)胞或組織后立即固定并且要充分固定,以阻止這些酶導(dǎo)致假陽(yáng)性。
2)使用了不適當(dāng)?shù)墓潭ㄒ?,例如一些酸性固定液,?dǎo)致出現(xiàn)假陽(yáng)性。建議采用新鮮配置的4%中性對(duì)聚甲醛固定液。
3)TUNEL檢測(cè)反應(yīng)時(shí)間過(guò)長(zhǎng),細(xì)胞或組織表面不能保持濕潤(rùn),也可能出現(xiàn)非特異性熒光。注意控制反應(yīng)時(shí)間,并確保TUNEL檢測(cè)反應(yīng)液能很好地覆蓋樣品。
Q:熒光背景很高?
1)高速分裂和增殖的細(xì)胞,轉(zhuǎn)錄水平高,有時(shí)也會(huì)出現(xiàn)細(xì)胞核中的DNA斷裂。
2)在激發(fā)光下長(zhǎng)時(shí)間的暴露,導(dǎo)致假陽(yáng)性的出現(xiàn)。
Q:標(biāo)記效率低?
1)使用乙醇或甲醇固定會(huì)導(dǎo)致標(biāo)記的效率較低。
2)固定時(shí)間過(guò)長(zhǎng),導(dǎo)致交聯(lián)程度過(guò)高。此時(shí)宜減少固定時(shí)間。
3)組織切片過(guò)厚,使得固定效果不理想,控制在10 μm以內(nèi)。
4)熒光淬滅。Fluorescence在普通光照10min就會(huì)嚴(yán)重淬滅。解決方法是需注意避光操作。
